Tabletkę EllaOne można przyjąć w dowolnym dniu cyklu. Można rozpocząć stosowanie antykoncepcji hormonalnej (wkładki domacicznej uwalniającej hormony) nawet od razu po przyjęciu tabletki awaryjnej, jednak w trakcie kolejnego cyklu należy stosować dodatkowe, mechaniczne metody antykoncepcji (np. prezerwatywa). W przypadku spirali niehormonalnej również nie ma przeciwwskazań.
