Stężenie metabolitu Amitryptyliny osiąga maksymalne stężenie w surowicy krwi w ciągu 8-24 h od przyjęcia doustnej dawki. Z tego wynika, że tabletka "działa" całą dobę, stąd Amitryptylinę dawkuje się raz dziennie.
