Zofenil jest prolekiem. Lek Zofenil, po przekształceniu w substancją czynną, osiąga maksymalne stężenie we krwi po upływie ok. 1,5 h od momentu zażycia tabletki. Okres półtrwania leku wynosi 5,5 h. Oznacza to, że po ok. 28 h stężenie substancji czynnej we krwi maleje praktycznie do zera.
